The next generation of drugs in development have tremendous potential to alleviate symptoms of once thought untreatable diseases. Many of these drugs require a delivery system along the lines of cell penetrating peptides. Cell penetrating peptides lack a foundational research component to determine the cellular uptake without vastly changing the environment of the bacterial cells. The development of a novel method to sense and relatively quantify cell penetrating peptide uptake in live cells will help continue the development of these next generation of drugs. With the addition of a fluorogenic labeled amino acid these peptides will only fluoresce inside cells. These cells can then be taken to a flow cytometer where a real time assay can be performed showing fluorescence which can be used to relatively quantify the uptake of these cell penetrating peptides into bacterial cells.
