Unveiling Cryptic Avifauna Diversity Using DNA Barcoding: A Case Study In the Passerine Genus Emberiza

Biodiversity loss poses a significant threat, requiring accurate assessment and conservation strategies. We developed a cost-effective molecular pipeline integrating DNA barcoding with Nanopore sequencing to study cryptic Emberiza (Passeriformes: Emberizidae) species. Using non-invasive feather DNA extractions from 242 specimens in Turkey's Aras River Riparian Corridor, we analyzed CO1 DNA barcodes. Phylogenetic results suggest cryptic species, emphasizing the region's conservation importance.
